Acute hepatotoxicity of Crotalus durissus terrificus (South American rattlesnake) venom in rats J. Venom. Anim. Toxins incl. Trop. Dis.
França,RF; Vieira,RP; Ferrari,EF; Souza,RA; Osorio,RAL; Prianti-Jr,ACG; Hyslop,S; Zamuner,SR; Cogo,JC; Ribeiro,W.
Venom of the South American rattlesnake, Crotalus durissus terrificus (Cdt), presents myotoxic and neurotoxic outcomes, but reports on its effects on the liver are scarce. This study examined the hepatotoxicity resulting from Cdt venom administration (100, 200 and 300 µg/kg) in male Wistar rats. Animals were studies at 3, 6, 9 and 12 hours after venom injection. The hepatotoxicity was assessed through serum levels of aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (AP), gamma glutamyl transferase (GGT), bilirrubin and also by histopathological evaluation. All the different concentrations of Cdt venom resulted in increased levels of hepatic enzymes, when compared with the control group, except for the 100 µg/kg dose,...

Antibacterial activity of venom from funnel web spider Agelena labyrinthica (Araneae: Agelenidae) J. Venom. Anim. Toxins incl. Trop. Dis.
Benli,M; Yigit,N.
Since the number of microorganisms that are resistant to antibiotics has been increasing steadily, the need for combating these pathogens requires new pharmaceutical agents. To produce these substances, new models have been developed in recent decades. In our study, the venom of Agelena labyrinthica (Clerck, 1757) (Araneae: Agelenidae) was tested against ten bacterial strains, specifically, testing 1/100, 1/10 and 1/1 fractions of diluted venom against these bacteria. While the 1/100 dilution was successful in only one of ten bacterial strains, the 1/10 and the 1/1 were effective on six of ten bacterial strains. The most effective results, among these three different concentrations, were observed on Bacillus subtilis. The other five strains that were also...

Antithrombotic effect of Lonomia obliqua caterpillar bristle extract on experimental venous thrombosis BJMBR
Prezoto,B.C.; Maffei,F.H.A.; Mattar,L.; Chudzinski-Tavassi,A.M.; Curi,P.R..
The venom of Lonomia obliqua caterpillar may induce a hemorrhagic syndrome in humans, and blood incoagulability by afibrinogenemia when intravenously injected in laboratory animals. The possible antithrombotic and thrombolytic activities of L. obliqua caterpillar bristle extract (LOCBE) were evaluated in this study. The minimal intravenous dose of the extract necessary to induce afibrinogenemia and anticoagulation was 3.0 and 10.0 µg protein/kg body weight for rabbits and rats, respectively. In rabbits, this dose induced total blood incoagulability for at least 10 h and did not reduce the weight of preformed venous thrombi, in contrast to streptokinase (30,000 IU/kg). In rats, pretreatment with 5.0 and 10.0 µg/kg LOCBE prevented the formation of thrombi...

Biodistribution studies of bee venom and spider toxin using radiotracers J. Venom. Anim. Toxins incl. Trop. Dis.
Yonamine,C. M.; Costa,H.; Silva,J. A. A.; Muramoto,E.; Rogero,J. R.; Troncone,L. R. P.; Camillo,M. A. P..
The use of radiotracers allows the understanding of the bioavailability process, biodistribution, and kinetics of any molecule labelled with an isotope, which does not alter the molecule's biological properties. In this work, technetium-99m and iodine-125 were chosen as radiotracers for biodistribution studies in mice using bee (Apis mellifera) venom and a toxin (PnTX2-6) from the Brazilian "armed" spider (Phoneutria nigriventer) venom. Incorporated radioactivity was measured in the blood, brain, heart, lung, liver, kidney, adrenal gland, spleen, stomach, testicle, intestine, muscle, and thyroid gland. Results provided the blood kinetic parameter, and different organs distribution rates.

Cardiotoxic effects of venoms from Chironex fleckeri and Chiropsalmus sp. on an invertebrate model J. Venom. Anim. Toxins incl. Trop. Dis.
Carrette,T.; Seymour,J..
Extracted venoms from two species of cubozoan, Chironex fleckeri (C. fleckeri)and Chiropsalmus sp., were injected into the freshwater crayfish Cherax quadricarinatus (C. quadricarinatus). By means of a Vascular Doppler, the cardiac parameters heart rate and contraction amplitude were recorded, with the theoretical total heart output calculated from these two recordings. Cherax quadricarinatus reflects prey items utilized by these two species in the wild and so comparisons of venom action on this animal have greater ecological relevance than those of previous mammalian models used. While the results of this study reflect previous research showing that the overall potency of C. fleckeri venom is greater than that of Chiropsalmus sp. venom, the action of the...

Characterization of insecticidal peptides from venom Australian funnel-web spiders J. Venom. Anim. Toxins incl. Trop. Dis.
Vonorax,E. J.; Tyler,M. I.; Atkinson,R. K.; Howden,M. E. H..
Australian funnel-web spiders are relatively large primitive hunting spiders. Male Atrax robustus spiders have been responsible for a number of human deaths. Venom was collected from the species Hadronyche infensa (Hickman) [female], H. formidabilis [male and female], H. versuta [female], and A. robustus (Cambridge) [male] and was fractionated by high performance liquid chromatography. This resulted in the isolation and purification of a homologous series of 7 insecticidal peptides of relatively low molecular mass (approximately 4kDa). The amino acid sequences of these toxins consisted of 36 or 37 amino acids and were named atracotoxins. For the major bioassay of these toxins, we used the cotton bollworm, Helicoverpa armigera (Hubner), due to the great...

Shirmardi,SP; Shamsaei,M; Gandomkar,M; Saniei,E; Ghannadi,M; Zare,A.
Iranian scorpions belong mainly to the Buthidae and Scorpionidae families, distributed into 16 genera and 25 species. In Iran, similar to other parts of the world, there are a few known species of scorpions responsible for severe envenoming; amongst which Mesobuthus eupeus is the most common. Its venom contains several toxin fractions that may affect the ion channel. In the present study purification, labeling and biological evaluation of M. eupeus venom are described. For separation, soluble venom was loaded on a chromatography column packed with Sephadex G-50 gel. Subsequently, the fractions were collected according to UV absorption at a wavelength of 280 nm. Toxic fraction (F3) was loaded on an anionic ion exchanger resin and then on a cationic resin....

Crotalus durissus terrificus crotapotin naturally displays preferred positions for amino acid substitutions J. Venom. Anim. Toxins incl. Trop. Dis.
Oliveira,Laudicéia Alves de; Ferreira Jr,Rui Seabra; Barraviera,Benedito; Carvalho,Francilene Capel Tavares de; Barros,Luciana Curtolo de; Santos,Lucilene Delazari dos; Pimenta,Daniel Carvalho.
Abstract Background Classically, Crotalus durissus terrificus (Cdt) venom can be described, according to chromatographic criteria, as a simple venom, composed of four major toxins, namely: gyroxin, crotamine, crotoxin and convulxin. Crotoxin is a non-covalent heterodimeric neurotoxin constituted of two subunits: an active phospholipase A2 and a chaperone protein, termed crotapotin. This molecule is composed of three peptide chains connected by seven disulfide bridges. Naturally occurring variants/isoforms of either crotoxin or crotapotin itself have already been reported. Methods The crude Cdt venom was separated by using RP-HPLC and the toxins were identified by mass spectrometry (MS). Crotapotin was purified, reduced and alkylated in order to...

Dose and time dependence of box jellyfish antivenom J. Venom. Anim. Toxins incl. Trop. Dis.
Andreosso,Athena; Smout,Michael J; Seymour,Jamie E.
Background The effectiveness of the currently available box jellyfish (Chironex fleckeri) antivenom has been subject of debate for many years. To assess whether the box jellyfish antivenom has the ability to attenuate venom-induced damage at cellular level, the present study analyzed the dose and time dependence of the antivenom in a cell-based assay. Methods Different doses of antivenom were added to venom and subsequently administered to cells and the cell index was measured using xCelligence Technology (ACEA Biosciences). Similarly, antivenom and venom were incubated over different time periods and cell survival measured as stated above. For both experiments, the cell index was plotted as a measure of cell survival against the dose or incubation time and...
